He was enrolled at the National Theatre School of Denmark at the age of 27. He graduated in 1995. He was also a part of The Killing, a drama show that was telecast across the globe. He later appeared in Borgen III and Those Who Kill, two internationally highly acclaimed Danish drama series. He also played the lead role in an episode of the BBC drama Sherlock as blackmailer Charles Augustus Magnussen. In the American TV show House of Cards, he played the role of the president of Russia. In 2018, he won an International Emmy Award for his part as the president of Russia on the Danish television drama Herrens Veje. It was shown in the UK in 2019. According to Mikkelsen, he was taught English from his older brother by watching Monty Python recordings and watching the comedy sketches.
